Background: Investigation for research on the cellular biology and physiology of colon cancer tissues requires viable dissociation single cells. The amount tissue methods can affect cell viability. Inadequate initial has negative effects data quality by resulting in insufficient number 
 Methods: In context this study different-weigh different-textured tumor have been evaluated to emphasize importance properties during operation processing isolation success. Necrotic areas were also with isolated cells success 3D primary culture. Results: Elevated weight resulted more total caused low percentage Since resected bigger than biopsy samples, derived culture succesfully maintained culture.
 Conclusion: To conclude, from non-necrotic showed better growth pattern cultures. On other hand, size was found as a crucial parameter obtaining
